Evaluation of Genetic Variability within a Collection of Cumin Genotypes Using RAPD, ISSR, SRAP and SCoT Markers and Variability of In Vitro Callus Induced Therefrom

The availability of adequate information about the documentation and characterization germplasm is fundamental for any crop improvement program. importance cumin as a medicinal plant yet lack its genetic variability encouraged us to initiate current study aiming at assessing among 17 genotypes from different geographical regions using four molecular markers (RAPD, ISSR, SRAP SCoT). Further, potential six accessions induce callus was studied under in vitro conditions on MS B5 basal media supplemented with various combinations between 2,4-D kinetin. Our findings showed that combining 87 primers, including 42, 15, 7 23 primers RAPD, SCoT SRAP, respectively, facilitated detecting relationship assessed accessions. A total number 765 bands were analyzed, which only 74 polymorphic. polymorphism low (9.67%) varied within markers. exposed highest average values content (0.06), resolving power (0.91) diversity index (0.08), while ISSR induced expected heterozygosity (0.06) marker (0.08). UPGMA dendrogram based data all separated into three main clusters, partial geographic-based genotypes. Out evaluated induction vitro, five potent callus, frequency ranging 90.4 97.5% no significant differences tested ANOVA. Two medium superior results: amended (4.44 mg/L) + Kin (0.22 (8.88 mg/L). Statistically variations relative growth rate produced detected accessions, where EG-4 values, followed by EG-5. All combinations, alone, exhibited superiority compared those both Kin. implying real need more effort assess wider populations geographic together reliable diversification programs.
